GHK-Cu Side Effects: What Is Documented, What Is Reported, and the Copper Question
The GHK-Cu safety story is usually told in one sentence: it is a naturally occurring human peptide, so it is safe. Half of that is true. GHK is a normal constituent of human plasma, and the cosmetic and wound care use of GHK-Cu goes back decades without a signal of harm. The other half is a leap, because naturally occurring says nothing about injecting milligrams of a copper complex subcutaneously every day, which is a use the human literature has never examined.

What the Human Evidence Actually Covers
There are two controlled human trials of GHK-Cu, and both are topical. Mulder and colleagues ran a multicentre, randomised, evaluator blinded, placebo controlled study of a GHK-Cu gel on diabetic neuropathic foot ulcers, framed explicitly as a safety and effectiveness study (Wound Repair Regen 1994, PMID 17147644). The treated ulcers closed faster and became infected less often, 7 percent against 34 percent for vehicle. The abstract reports no adverse event data one way or the other, which for a daily topical on open wounds in diabetics is a quiet form of reassurance, not proof.
The second is smaller. Miller and colleagues randomised patients to a GHK-Cu skin care regimen or the same regimen without it after CO2 laser resurfacing around the mouth, and in the 13 who completed it blinded evaluators found no difference in erythema resolution and no objective difference in wrinkles or skin quality at 12 weeks, with only patient satisfaction higher in the GHK-Cu group (Arch Facial Plast Surg 2006, PMID 16847171). Its abstract, like Mulder's, reports no adverse event data either way, though a copper peptide product on freshly resurfaced skin is the closer test of topical tolerability for a page like this one.
Beyond that, the topical safety record is the cosmetic industry's. GHK-Cu has been sold in skincare under the INCI name Copper Tripeptide-1 for decades, and the reviews by its discoverer describe controlled studies on aged skin (Pickart, J Biomater Sci Polym Ed 2008, PMID 18644225). Those studies are mostly conference abstracts and company data rather than indexed trials, so the absence of reported harm in them is real but lightly documented.
For GHK-Cu on its own by injection, there is nothing: no human trial, no dose finding study, no pharmacokinetic study. The one published human series of injected GHK-Cu used it as one ingredient of a six component growth factor cocktail injected intradermally into the scalp, open label and uncontrolled, in 1,000 patients (Kapoor and Shome, J Cosmet Laser Ther 2018, PMID 29482481); safety was followed by physical examination, questionnaires and laboratory tests, the treatment was reported as well tolerated, and the paper says nothing about GHK-Cu alone or about subcutaneous systemic use. Every statement about the tolerability of the subcutaneous route, including the ones below, is anecdote or inference from animals. The frequency language is deliberately qualitative. Nobody has counted, and a page that gives you percentages for the injected route is inventing them.
The injection sting
GHK-Cu is widely reported to burn on injection, which is not a common complaint with most research peptides, and it is the single most common complaint about this one. There is no published explanation. The plausible candidates, the copper ion, the pH of the reconstituted solution and the concentration, have not been tested against each other. What people do about it is dilute, so the dose sits in a larger volume at a lower concentration, and inject slowly. The blue tint
Copper(II) complexes are blue to violet, GHK-Cu solution is visibly so, and a small volume sitting in subcutaneous fat shows through pale skin for a few hours. It is not a bruise, it is not a reaction, and it disperses as the solution does. It is worth saying once because it alarms people the first time.
Fatigue, headache and mood
A minority of people injecting GHK-Cu report feeling flat, tired or headachy in the first days of a block, and a smaller number report the opposite, a lift in mood. The Bureau's guide mentions both. There is no established mechanism for either and no incidence data. No published study describes it. The candidates are the formulation rather than the peptide, a clash between the copper complex and the vitamin C or acids in the same routine, and simple over use. This is the only part of GHK-Cu safety that is specific to the molecule, and it is arithmetic rather than opinion. GHK-Cu is about 16 percent copper by mass: the tripeptide weighs roughly 340 daltons, the copper ion roughly 64. A 1 mg injection carries about 0.16 mg of copper, a 2 mg injection about 0.3 mg, and an eight week block at 2 mg daily delivers on the order of 18 mg of copper in total.

On the face of it a 2 mg injection adds a third of a day's dietary copper and about a thirtieth of the upper limit, which is a comfortable margin. Two things narrow it. The dietary figures assume oral intake, where the intestine regulates how much copper is absorbed and the liver disposes of the excess in bile; an injection skips the first step entirely. And the upper limit was set to avoid liver damage from chronic excess, not to describe a level at which nothing else happens. Neither point makes 0.3 mg dangerous. Both are reasons the cycle breaks in circulation are sensible for a compound that has never been studied long term in people.
A separate copper issue is the product itself. GHK binds copper at a defined one to one ratio, and a vendor can sell free GHK with too little copper, or GHK with copper in excess, and label both as GHK-Cu. Who Should Not Use GHK-Cu
- Anyone with Wilson's disease or another disorder of copper handling. Wilson's disease is a genetic failure to excrete copper, treated with copper chelators and a low copper diet. Adding copper by any route, including topical, works directly against the treatment. This is the one absolute.
- Anyone on a copper chelator such as penicillamine or trientine, for the same reason.
- Pregnancy and breastfeeding. No data by any route. The default for an unstudied compound applies.
- Active cancer or a recent history of it. GHK-Cu increases vascular endothelial growth factor and stimulates angiogenesis and cell migration in wound models (Pickart 2008, PMID 18644225), which are processes a tumour also depends on. Against that, a Connectivity Map analysis predicted that GHK would reverse a metastasis-prone gene signature identified in early stage colorectal cancer, a computational prediction rather than a treatment result (Hong et al., Clin Exp Metastasis 2010, PMID 20143136) and the discoverer's reviews describe anti-cancer actions. Nobody has resolved this in a person. It is a question for the oncologist, not a forum.
- Known copper contact allergy, for the topical route.
- Long injected blocks with no liver monitoring deserve more caution than short ones. Not because harm is expected, but because copper is cleared by the liver and the block is unstudied.
A copper IUD comes up often. There is no data and no mechanistic reason to expect an interaction; the device releases copper locally in the uterus and that is unrelated to a subcutaneous dose. It belongs on the list of things nobody has studied, not the list of things to worry about.
What Is Not a Side Effect
Two things get blamed on GHK-Cu that are not the peptide. The first is the general injection site reaction, redness, a small lump, a bruise, which is technique and site rotation and is covered in the injection guide.
