Topical GHK-Cu: Serums, Solutions, and Why the Percentage Is Not the Point
Topical GHK-Cu is the one form of the compound with human trial data behind it, and it is also the form where the number on the label tells you least. A copper peptide serum can say 1 percent, a research solution can say 2 percent, a scalp tonic can say 0.1 percent, and the mechanism data suggests that all three are overkill by a factor of millions at the point where the peptide acts. What separates a topical that does something from one that does not is whether the peptide gets through the skin, and that is a delivery question the percentage does not answer.

What Topical GHK-Cu Is
GHK-Cu is the tripeptide glycyl-L-histidyl-L-lysine bound to a copper(II) ion. It occurs naturally in human plasma, saliva and urine and its level falls with age (Pickart et al., Biomed Res Int 2015, PMID 26236730). In cosmetics it is listed as Copper Tripeptide-1, and in the four way classification of topical peptides used in Gorouhi and Maibach's review, signal, enzyme inhibitor, neurotransmitter inhibitor and carrier, copper peptides sit in the carrier group, the one whose job is to deliver a trace element, as distinct from signal peptides like Matrixyl and neurotransmitter inhibiting peptides like Argireline (Int J Cosmet Sci 2009, PMID 19570099).
GHK-Cu is a carrier, but it is also the one topical peptide among those categories with a plausible claim to broad gene regulation: the discoverer's reviews report that it up or down regulates several thousand human genes in expression datasets (Pickart et al. 2015), and independent groups querying the Connectivity Map have found GHK predicted to reverse the gene expression signature of emphysema, a prediction that held up when human lung fibroblasts were treated with it (Campbell et al., Genome Med 2012, PMID 22937864), and predicted to reverse a metastasis-prone signature in colorectal cancer (Hong et al., Clin Exp Metastasis 2010, PMID 20143136). Whether any of that happens in skin under a serum is a different question, and the answer runs through penetration.

Maquart and colleagues showed in 1988 that GHK-Cu stimulates collagen synthesis in cultured fibroblasts from a concentration of about a picomolar, with the maximum effect at one nanomolar (FEBS Lett, PMID 3169264). One nanomolar is about 0.4 micrograms per litre. A 1 percent solution is 10 grams per litre, roughly 25 millimolar, which is twenty five million times that concentration. Even a 0.1 percent scalp tonic is two and a half million times it.
Two things follow. The first is that the label percentage is not a measure of potency at the target. Cells respond to nanomolar GHK-Cu, and any product that delivers a tiny fraction of its content into the dermis is delivering more than enough. The second is the real variable: how much gets in. GHK-Cu weighs about 404 daltons, under the 500 dalton rule of thumb for passive diffusion through the stratum corneum, but it is hydrophilic and charged, which is the wrong profile for crossing a lipid barrier. Mazurowska and Mojski measured copper peptide complexes crossing a liposome membrane built to mimic the stratum corneum lipids in a Franz diffusion cell, and found that they do permeate and that the peptide changes how the copper moves (J Cosmet Sci 2008, PMID 18350235). That is a model membrane, not skin, and it establishes that penetration is possible, not how much.
The questions that matter for a topical are therefore the vehicle, whether the barrier has been disrupted, and how long the product sits on the skin. The label percentage is at best a proxy for whether the formulator was serious.

The strongest human data for topical GHK-Cu is a wound trial. Mulder and colleagues' multicentre randomised, placebo controlled study of a GHK-Cu gel on diabetic foot ulcers reported a median 98.5 percent closure against 60.8 percent for vehicle, closure three times faster, and 7 percent infections against 34 percent (Wound Repair Regen 1994, PMID 17147644). Rat work with the same gel found a 2 percent formulation closed ischaemic wounds faster, with lower TNF-alpha and lower matrix metalloproteinase 2 and 9 in the tissue (Canapp et al., Vet Surg 2003, PMID 14648529), and an earlier rat model showed the compound modulating which metalloproteinases were expressed during remodelling (Siméon et al., J Invest Dermatol 1999, PMID 10383745). Injured skin, where the barrier is gone, is where topical GHK-Cu is best established.
Intact aged skin is a different case. The reviews by the discoverer describe controlled studies in which topical GHK-Cu tightened skin, improved elasticity and firmness, reduced fine lines and photodamage and increased keratinocyte proliferation (Pickart, J Biomater Sci Polym Ed 2008, PMID 18644225). The underlying studies are mostly conference abstracts and industry data from the late 1990s and early 2000s rather than indexed trials, the reviews are written by the founder of a company that sells copper peptide products, and the one independent randomised trial that scored wrinkles, on laser-resurfaced skin rather than intact skin, found no objective difference: Miller and colleagues randomised patients to a GHK-Cu skin care regimen or the same regimen without it after CO2 laser resurfacing around the mouth, and in the 13 who completed it blinded evaluators saw no difference in erythema resolution, wrinkles or overall skin quality at 12 weeks, with only patient satisfaction higher in the GHK-Cu group (Arch Facial Plast Surg 2006, PMID 16847171). The mechanism work is solid: GHK-Cu suppresses inflammatory IL-6 secretion in human dermal fibroblasts (Gruchlik et al., Acta Pol Pharm 2012, PMID 23285694) and drives collagen, elastin and glycosaminoglycan synthesis in culture (Pickart and Margolina, Int J Mol Sci 2018, PMID 29986520). The honest expectation, then, is the one the guide gives: texture and tone in the second and third month, firmness later, judged by photographs in the same light rather than by impression. That is the same timescale as the collagen support peptides in Matrixyl 3000, and slower than the expression line peptides such as Argireline, which plateau at four to eight weeks.
What the Evidence Supports, Hair
The hair data is preclinical and it is worth being precise about what was tested. Pyo and colleagues showed that a copper tripeptide stimulated elongation of human hair follicles ex vivo and proliferation of dermal papilla cells at picomolar to nanomolar concentrations (Arch Pharm Res 2007, PMID 17703734). The peptide in that study was AHK-Cu, alanyl-histidyl-lysine copper, a close relative of GHK-Cu and not the same molecule. Uno and Kurata, reviewing their primate alopecia work, reported that a copper binding peptide coded PC1031 enlarged follicles on the back skin of fuzzy rats with an effect similar to topical minoxidil (J Invest Dermatol 1993, PMID 8326148); the abstract identifies it only by that code, not as GHK-Cu. There is no human trial of topical GHK-Cu for hair loss on PubMed; the closest human report is an uncontrolled 1,000 patient series of intradermal scalp injections of a growth factor cocktail that contained copper tripeptide-1 (Kapoor and Shome, J Cosmet Laser Ther 2018, PMID 29482481), which is neither topical nor GHK-Cu alone.
That does not make the hair use unreasonable. The mechanisms overlap, the follicle sits in the dermis where wound healing signals act, and the hair loss page sets out the 0.1 to 0.5 percent scalp protocols in circulation and why microneedling is so often paired with them. It does mean that "GHK-Cu regrows hair" is a claim about related copper peptides in rats and follicles in a dish, and the expectation should be sized accordingly: months, modest, and alongside the treatments that do have trial data rather than instead of them.
Reading a Label
- Look for Copper Tripeptide-1 in the ingredient list, and where it sits. Near the top means a meaningful amount; after the preservative means a trace.
- A stated percentage is better than none, but read what it is a percentage of. Some labels quote a peptide blend or a supplier solution rather than GHK-Cu itself, the same trick the Argireline page describes.
- Colour is a rough proxy. GHK-Cu solution is blue to violet at the strengths research vendors sell. A product that is colourless contains either very little or a different copper source. It is a proxy, not a measurement.
- Water based beats heavy cream for a hydrophilic peptide.

What follows describes how a finished cosmetic serum, the Copper Tripeptide-1 product on a skincare shelf, is used in the cosmetic studies and in practice; it is a description, not an instruction, and a solution mixed from a research vial is a research preparation, covered in the next section on that footing.
- Clean, dry skin, early in the order. It belongs in the water phase, before oils and occlusives.
- Once or twice daily. The guide describes both; consistency matters more than which.
- Keep it away from vitamin C and direct acids in the same step.
