Retatrutide is the newest of the three major weight loss peptides. It's still in Phase 3 clinical trials but shows the most impressive preliminary results. The triple mechanism (GLP-1, GIP, and glucagon activation) targets appetite, nutrient absorption, and metabolism simultaneously.
Retatrutide's Phase 2 obesity trial (Jastreboff et al., NEJM 2023) used four target maintenance doses, with stepwise titration in the higher-dose arms:
Trial results at 48 weeks: approximately 8% mean weight loss on 1 mg, 17% on 4 mg, 23% on 8 mg, and 24% on 12 mg. Retatrutide is currently in Phase 3 (the TRIUMPH program); FDA approval is most likely 2027 or 2028. As of April 2026 it is not yet approved.
Timeline with retatrutide: Similar to tirzepatide but potentially steeper in months 2-3. Most users report faster initial results due to the triple mechanism. However, more pronounced side effects are common, especially at dose escalations.
