Almost every page about copper peptides for hair cites two studies, and almost every one of them attributes both to the wrong molecule. The follicle elongation experiment used AHK-Cu. The rat study used a peptide the abstract calls PC1031. Neither is GHK-Cu, which is the copper peptide people actually buy, and which has no hair study of its own in any species.
That is not a reason to dismiss the category. It is a reason to know what you are buying. GHK-Cu and AHK-Cu Are Not the Same Molecule
Both are copper complexes of a three amino acid peptide ending in histidine and lysine, and the histidine is where the copper sits. The only structural difference is the first residue: glycine in GHK, alanine in AHK, a single extra methyl group and about 14 daltons. Chemically they are siblings. In evidence terms they are opposites.
GHK-Cu is the famous one. It was identified in human albumin in 1973, its plasma level falls with age (Pickart et al., Biomed Res Int 2015, PMID 26236730), it has a randomised controlled trial in diabetic foot ulcers behind it (Mulder et al., Wound Repair Regen 1994, PMID 17147644), and it has a large mechanism literature. Every vendor on the scorecard carries it, and its hair claim is entirely borrowed. AHK-Cu is the obscure one, with almost no skin literature and the only direct human follicle experiment in the category, carried by two vendors we score. Pyo and colleagues, 2007. A copper tripeptide stimulated elongation of human hair follicles cultured outside the body and made dermal papilla cells proliferate, at picomolar to nanomolar concentrations (Arch Pharm Res, PMID 17703734). The peptide was AHK-Cu. This is the only experiment in the category that put a copper peptide on real human follicles and measured them growing, and it is the reason AHK-Cu exists as a product at all. Its limit is obvious: a follicle in a dish has no stratum corneum over it, no blood supply and no androgen environment, so it says what the molecule can do, not what a serum will do.
Uno and Kurata, 1993. A copper binding peptide identified only by the code PC1031 enlarged follicles on the back skin of fuzzy rats, with an effect the authors compared to topical minoxidil (J Invest Dermatol, PMID 8326148). Follicle enlargement is the right endpoint, because a bigger follicle makes a thicker shaft, which is what miniaturisation takes away. The problem is the identification: the abstract never says PC1031 is GHK-Cu, and the hundreds of pages that cite it as a GHK-Cu result are asserting something the paper does not.
Kapoor and Shome, 2018. An open label, uncontrolled series of 1,000 patients given intradermal scalp injections of a six component growth factor cocktail, one ingredient of which was copper tripeptide-1 (J Cosmet Laser Ther, PMID 29482481). With no control arm and five other actives in the syringe it cannot attribute anything to the copper peptide, but it is the only human scalp report that exists.
The mechanism underneath all of this is the solid part. GHK-Cu raises VEGF, which builds the capillary loop that feeds a follicle, and KGF and FGF-2, which drive keratinocyte proliferation, and fibroblasts respond to it from about a picomolar, peaking near one nanomolar (Maquart et al., FEBS Lett 1988, PMID 3169264). A follicle is a dermal appendage surrounded by exactly the tissue that literature describes. The gap is not plausibility; it is that nobody has run the trial, and the GHK-Cu guide sets out the same gap for skin. Scalp solutions in circulation run 0.1 to 0.5 percent, against the 1 to 2 percent used on the face. The reason usually given is coverage rather than potency: a scalp takes more millilitres than a cheek. Since cells respond at a nanomolar and 0.1 percent is already millions of times that, the lower strength costs nothing that matters, as the topical GHK-Cu page works through in detail.
Mixing is arithmetic. Percent is milligrams per millilitre divided by ten, so 50mg of powder in 50 mL of bacteriostatic water is 0.1 percent and the same vial in 10 mL is 0.5 percent. A research solution in bacteriostatic water has no emollients, no penetration enhancer, no pH buffering and no stability testing. Injected Into the Scalp, and What Documents It
Two injected practices exist and they are not the same thing. Clinic mesotherapy places small amounts intradermally across the scalp in a grid, usually monthly, and the figures in circulation are 0.1 to 0.2 mg per site. Its only human documentation is the uncontrolled cocktail series above, it is a procedure done by someone who does it professionally, and the Bureau does not describe a home version of it.
Subcutaneous injection into the abdomen is the other, and it is systemic: 1 to 2 mg, three times weekly to daily, in blocks of eight to twelve weeks when the goal is hair rather than skin, because the follicle cycle is slower than skin turnover. Those amounts are a convention rather than a measured dose, and the copper arithmetic matters here in a way it never does with a serum. GHK-Cu is about 16 percent copper by mass, so 2 mg carries roughly 0.3 mg of copper, about a third of the adult dietary reference intake, and injected copper bypasses the intestinal step that regulates absorption.
